SARS COV 2 DIAGNOSIS USING RT-PCR AND CHROMATOGRAPHY METHOD
SARS COV 2 "OR" COVID-19 is a virus that is originated from mammals and then transmitted into humans that's why also known as Zoonotic Virus. Corona Virus Belongs to Coronaviridae family and has Positive (+ve) Strands of RNA there are several complications still existed in the diagnosis of Coronavirus. RT-PCR is more efficient, accurate and reliable than Chromatography Method also known as Gold Plated Antigen Kit Based Method.
PCR (Polymerase Chain Reaction):
There are three main stages in PCR in Each Cycle
1. Denaturation (94-98 Degree Celsius) For 1 Minute.
In this stage double strands of DNA are separated from each other.
2. Annealing (50-68 Degree Celsius) For 45 Seconds.
In this stage Each Separated stand binds with primer either forward or reverse primer. Specifically 3' to 5' nucleotide Or 5' to 3' nucleotide.
Example:
5' to 3' Nucleotide.
TGGT
3' to 5' Nucleotide.
ACCA
3.Elongation (70-75 Degree Celsius) For 1-2 Minutes.
In this stage nucleotides bind to specific site of primer to make another complete single stand.
Note:
All these steps were repeated in thermocycler. After 10 cycles 1024 copies of stands were formed, and after 20 cycles about 1 million (10 Lac) Copies were formed and after 30 cycle more than a billions copies of single stand were formed.
RT PCR:
RT-PCR stands for (Reverse Transcription - Polymerase Chain Reaction) is a fluorescent based spectrophotometry technique used for qualitative analysis. RT-PCR is a widely usable technique for the detection of SARS-COV-2 this technique convert RNA (Ribonucleic Acid) into cDNA (Complementary Deoxyribonucleic Acid) by the help of an enzyme known as Reverse Transcriptase.
